ratio nal anti-c ancer drug therapies research efforts are co ncentrated on unravelling the molecular ci rcuits whic h induce programmed cell death and bloc k proliferatio n of cancer cells.Mo dern therapeutic st rategies are based on the underst anding of the co mplexit y of physio logic al functio ns such as differenti atio n,development,immune responses,cell-c ycle arrest,DNA damage repair,apoptosis,auto phagy,energy metabolis m,and senesc ence.It has beco me evident that this knowledge will provide the means to target the co mponents of the pathways involved in these processes in a specific and selective manner thus paving the way for the development of effecti ve and perso nalis ed anti-c ancer therapies.Transc riptio n is a cruci al cellular proc ess that regulates a multit ude of physio logic al functio ns,whic h are ess enti al in dis ease progressio n and cellular response to therapy.Transc riptio n facto rs such as the p53 tumor suppresso r and the hypoxia-i nduci ble facto r-α(HIF-α)are key players in carci nogenesis and cellular response to cancer therapies.Bot h of these transc riptio n facto rs regulate gene expressio n of genes involved in cell death and proliferatio n,in so me cases coo perati ng to wards produci ng the same outco me and in so me ot hers mediati ng opposi ng effects.It is thus apparent that fine tuning of the acti vit y of these transc riptio n facto rs is ess enti al to determine the cellular response to therapeutic regimens,in ot her words whether tumor cells will co mmit to apoptosis or evade engagement wit h the anti-proliferati ve effects of drugs leading to drug resist ance.Our observatio ns support the notio n that the functio nal crosst alk between HIF-1αand p53 pathways and thus the fine tuning of their transcriptio nal acti vit y is mediated by co facto rs shared between the two transc riptio n facto rs such as co mponents of the p300 co-acti vato r multi prot ein co mplex.In partic ular,there is evidence to suggest that differenti al co mpositio n of the co-modulato
